<p>?Before this, the only way to tell if genome engineering occurred was to do a forensic analysis,? said Paul Abraham, a bioanalytical chemist and head of ORNL?s Secure Ecosystem Engineering and Design Science Focus Area. ?To be successful, you would need to know what the genome looked like before it was rewritten. We wanted to design a platform where we could proactively observe CRISPR activity.?</p>
<p>The research team developed an efficient self-detect solution that takes advantage of the way CRISPR works to trigger the technology to reveal itself. Under normal conditions, CRISPR works by connecting with a short RNA sequence, known as the guide RNA, as it leads CRISPR to a matching DNA sequence. When the target DNA is found, CRISPR modifies the DNA by acting like tiny molecular scissors to cut through one or both strands of DNA, depending on the type of CRISPR technology in use.</p>
<p>Abraham likens their method to an alarm system with two components: a biosensor guide RNA that redirects CRISPR activity and a reporter protein that flags the activity. Researchers encode the two components into an organism?s DNA to enable the monitoring system.</p>
<p>With the self-detect system in place, the biosensor guide RNA intercepts CRISPR, preventing CRISPR from connecting with its original gene target and redirecting CRISPR to a specific DNA sequence that encodes for a nonfunctioning green fluorescent protein, or GFP. When CRISPR edits the sequence, it flips a switch that produces functioning GFP, which creates a green glow signaling CRISPR?s presence.</p>
<p>Because a microscope is required to see the glow from GFP, the researchers improved on their original method by replacing GFP with a similar reporter protein, called eYGFPuv, that is visible under the type of ultraviolet light commonly known as a black light.</p>
<p>?Now we can see whether CRISPR is active in real time regardless of the size, shape and location of the organisms we?re evaluating,? Abraham said. ?This flexibility speeds the bioengineering process and extends the biosensors? use in laboratory and field applications.?</p>
<p>Since CRISPR must be tailored to each organism for effective use, knowing whether the CRISPR technology is working in a particular plant or microbe can accelerate progress toward goals such as developing drought-resistant bioenergy crops and engineering bacteria to efficiently convert plants into sustainable aviation fuels.</p>
<p>?These tools allow us to quickly identify positive transformants with desired genetic changes we are targeting,? said Carrie Eckert, lead for ORNL?s Synthetic Biology Group. ?We are easily able to see the variants versus those where modification did not occur.?</p>

They demonstrated the sensors individually with each sensor signaling the presence of a particular type of CRISPR tool. Abraham also sees the potential to combine the biosensors into a version that would flag multiple gene editing technologies at once.</p>

<p>The study published today in Nature Communications outlines the genes that are switched on or off in osteocytes, a type of bone cell that controls how other types of cells make or break down parts of the skeleton to maintain strong and healthy bones.</p>
<p>?This new information provides a kind of genetic shortlist we can look to when diagnosing bone diseases that have a genetic component,? says the study?s first author Dr Scott Youlten, Research Officer in the Bone Biology Lab. ?Identifying this unique genetic pattern will also help us find new therapies for bone disease and better understand the impacts of current therapies on the skeleton.?</p>
<p><strong>A first look at the osteocyte transcriptome </strong></p>
<p>The skeleton is a highly dynamic structure that changes shape and composition throughout a person?s life. Osteocytes are the most abundant cell type in bone but have proved difficult to study because they are embedded within the hard mineral structure of the skeleton.</p>
<p>Inside the bone, osteocytes form a network similar in scale and complexity to the neurons in the brain (with over 23 trillion connections between 42 billion osteocytes) that monitors bone health and responds to ageing and damage by signalling other cells to build more bone or break down old bone. Diseases such as osteoporosis and rare genetic skeletal disorders arise from an imbalance in these processes.</p>
<p>To understand what genes are involved in controlling bone build-up or breakdown, the researchers isolated bone samples from different skeletal sites of experimental models to measure the average gene activity in osteocytes. Through this, they mapped a comprehensive osteocyte ?signature? of 1239 genes that are switched on in osteocytes and that distinguish them from other cells. 77% of these genes have no previously known role in the skeleton and many were completely novel and only found in these critical cells.</p>
<p>?Many of the genes we saw enriched in osteocytes are also found in neurons, which is interesting given these cells share similar physical characteristics and may suggest they are more closely related than we previously thought,? explains Dr Youlten.</p>
<p>A comparison of the osteocyte signature genes with human genetic association studies of osteoporosis identify new genes that maybe associated with susceptibility to this common skeleton disease. Furthermore many of these osteocyte genes were also shown to cause rare bone diseases.</p>
<p>?Mapping the osteocyte transcriptome could help clinicians and researchers more easily establish whether a rare bone disease has a genetic cause, by looking through the ?shortlist? of genes known to play an active role in controlling the skeleton,? says Dr Youlten.</p>
<p><strong>The big picture view </strong></p>
<p>Co-senior author Professor Peter Croucher, Deputy Director of the Garvan Institute and Head of the Bone Biology Lab, says that ?the osteocyte transcriptome map gives researchers a picture of the whole landscape of genes that are switched on in osteocytes for the first time, rather than just a small glimpse.?</p>
<p>?The majority of genes that we?ve found to be active within osteocytes had no previously known role in bones. This discovery will help us understand what controls the skeleton, which genes are important in rare and common skeletal diseases and help us identify new treatments that can stop development of bone disease and also restore lost bone.?</p>

Through this, they mapped a comprehensive osteocyte ?signature? of 1239 genes that are switched on in osteocytes and that distinguish them from other cells. 77% of these genes have no previously known role in the skeleton and many were completely novel and only found in these critical cells.</p>
<p>?Many of the genes we saw enriched in osteocytes are also found in neurons, which is interesting given these cells share similar physical characteristics and may suggest they are more closely related than we previously thought,? explains Dr Youlten.</p>
<p>A comparison of the osteocyte signature genes with human genetic association studies of osteoporosis identify new genes that maybe associated with susceptibility to this common skeleton disease. Furthermore many of these osteocyte genes were also shown to cause rare bone diseases.</p>
<p>?Mapping the osteocyte transcriptome could help clinicians and researchers more easily establish whether a rare bone disease has a genetic cause, by looking through the ?shortlist? of genes known to play an active role in controlling the skeleton,? says Dr Youlten.</p>
<p><strong>The big picture view </strong></p>
<p>Co-senior author Professor Peter Croucher, Deputy Director of the Garvan Institute and Head of the Bone Biology Lab, says that ?the osteocyte transcriptome map gives researchers a picture of the whole landscape of genes that are switched on in osteocytes for the first time, rather than just a small glimpse.?</p>
<p>?The majority of genes that we?ve found to be active within osteocytes had no previously known role in bones. This discovery will help us understand what controls the skeleton, which genes are important in rare and common skeletal diseases and help us identify new treatments that can stop development of bone disease and also restore lost bone.?</p>

The Magic in the first half played better than the Lakers, and without the sensational blackout of the third period they probably would have played until the end.  But for a young group that looks to tomorrow rather than today, even in a defeat like this there are important lessons. </p>

Saturday 18 December at 2 pm in Parma against Romania, the third in two months of this new route after Spain and Uruguay.  To tell it, and to take stock of the tournament after the first 10 days, is Andrea Marcato.  Coach of Petrarca Padova, unbeaten winter champion and assistant to coach Alessandro Troncon of the second national team (this week due to indisposition he will be replaced by Gonzalo Garcia).</p>

Thanks to what happened at the Tokyo Olympics at the beginning of August, an edition that, as a long wave of emotions, seems to never end.  Trento will have the merit and the honor of bringing together the seven athletes who have won five historic gold medals in Japan.  Yes, five.  Two more than the maximum blue, which dated back to Moscow 1980 and Los Angeles 1984, the Games, however, halved by boycotts.  In short: an Olympics like this, who knows when it will happen again.</p>

The policeman will be accompanied by an exceptional guest, Valerij Borzov, the former Soviet now Ukrainian, bitter rival of Pietro Mennea, who at the 1972 Munich Games was gold in the 100 and 200. The two will meet again later the awards ceremony at the New Tokyo National Stadium.  It was last August 2nd and Valerij, in the role of IOC member, handed Marcell his wonderful gold medal (topped off by the European record of 9 ?80).  Immediately after (the scenario will not change), however <i class="italic">Blue Lightning</i>, together with Paolo Camossi&#8217;s pupil, on stage there will be the three companions of the 4&#215;100 of wonders: Lorenzo Patta, Fausto Desalu and Filippo Tortu. Everyone wants to be the first to build a metaverse, own more brands, and have as much power in their hands as possible over all the works of art that people care about.  It&#8217;s not just Epic Games &#8211; almost everyone wants a slice.</p>

<p>Just this week, Epic Games announced that it has acquired Harmonix, the developer known for Rock Band.  Harmonix &#8220;will collaborate closely with Epic to develop musical journeys and gameplay for Fortnite&#8221;, according to a note from Epic, which means that we will probably not see more Harmonix rhythm games: they will be only a part of Fortnite from now on.  Epic Games also owns Psyonix, the creators of Rocket League, and Tonic Games Group, the home behind Fall Guys.  And rest assured that others will follow.</p>
<p>?It&#8217;s no secret that Epic is interested in building the metaverse, and Tonic Games shares that goal,? Epic Games founder and CEO Tim Sweeney said at the time.  &#8220;Epic works to build this virtual future, but we need great creative talent who know how to create powerful games, content and experiences.&#8221; </p>

<p>?Contents and experiences?: this is the important part of the declaration.  This is the real plan.  Epic Games will continue to incorporate studies to power everything Fortnite can do.  Consolidation (the practice whereby large fish eat small fish, becoming bigger and bigger and, in fact, solid) is the enemy of diversity.  The goal is <b>make all games, <i>everything</i>, Fortnite.</b> We&#8217;re already seeing commercials for Nike Jordan, John Wick, and Nfl popping up in the game, and we&#8217;re getting closer to a dystopian world that looks like it&#8217;s straight out of Blade Runner.</p>

<h2 class="is-h5 is-inline is-uppercase is-wp-float">Why is it a problem &#8211;</h2>
<p>The fact is that in some ways we already live in the metaverse.  Log into Twitter on your phone and you&#8217;re in another world full of people you probably won&#8217;t interact with once you&#8217;re out of it.  Twitter has made people take jobs, get in trouble, and meet a partner.  The web is already impacting our lives in real and tangible ways.  The idea of ??metaverse video games is just another way to &#8220;entertain&#8221; people for money.  In the last few years, <b>?Engagement? was a key metric</b> for the success of video game companies.  But was it a metric that did gamers good?  No not at all.</p>

<p>The fact that players need to be constantly entertained in their video games is what has led to the creation of games as a service, battle passes and open world games where you play the game. <b>same exact activity 5,000 times.</b> It is what gave the loot boxes and all the similar mechanics, which have the sole objective of making us continue playing, and the metaverse is only an extension of it.  And this path will lead to a situation where fewer smaller creators will be able to do the things they want to do, and more giant games and experiences that require all of your free time, luring you in with constant events and limited-time offers to keep you coming back.</p>
<p>This is not good for the developers either.  Sure, consolidation helps some struggling teams stay afloat in the same way it does working externally, but do developers really want to work on the same project forever?  It sounds difficult, and this will ultimately lead to creative burnout and the death of talent.  After all, if you don&#8217;t like working in video games, why not try the much more lucrative route of software development?  The events of recent times show <b>a glimpse of the future</b>, and what we have seen so far is certainly not exciting.</p>

<p>According to the tweet, Sonic is playable in Fortnite.  It&#8217;s not a skin, it&#8217;s a whole game, the new Sonic Origins.  Epic Games owns Sega in 2030, along with a handful of other huge mid-tier publishers and developers. <b>Microsoft, Sony, Embracer Group, EA and Tencent</b> all the others possess.</p>
<p>Out of pure curiosity, start Fortnite and find out what it has become <b>come Ready Player One:</b> an entirely digital world in which to play, meet friends, have fun? in one word, live as in an alternative reality.  There is no need for a console, as it comes pre-installed on all TVs, eye implants and VR headsets.  Head to Fortnite Plaza, a hub area where all your friends and family are enjoying their free time.  Your wife is here, although she hates video games, because Epic has a deal with the big supermarkets where you can get a big discount available only for those who shop in the metaverse.  He is strolling through the virtual shelves of a virtual store and filling a shopping cart, something Epic began to master when it implemented the shopping cart functionality in the Epic Games Store in 2025.</p>
